Deon began his comedy career in Chicago. He wrote for Conan’s shows The Tonight Show with Conan O'Brien (from 2009 to 2010) and Conan (from 2010 to 2012). He made a breakthrough with his role as Charlie Telphy in the ABC sitcom Black-ish and won multiple awards for his role. He has also appeared on the shows Angie Tribeca and Grown-ish. He began his own show, Deon Cole's Black Box, in 2013, under his own production house. However, the show was canceled soon after. In 2019, he was featured in the Netflix special Deon Cole: Cole Hearted. He has been seen in films such as the Barbershop series and Holiday Rush. In 2020, he released a video titled 40 Minutes of Deon Cole "Workin' It Out” on YouTube, which contained clippings of his practice sessions for his 2020 comedy tour that was canceled due to COVID-19. He is extremely secretive about his personal life, but it is known that he has a son.

TriviaDeon Cole once recalled a funny incident that occurred at a Santana concert that featured singer Stevie Wonder. Apparently, Deon and his then-girlfriend met Stevie backstage. Following this, Deon left, leaving his girlfriend alone with Stevie. When he went back after an hour, Deon’s girlfriend told him that she would be staying back to console Stevie about a few things he was upset with. She also paid him $100 and asked him to go back home.
